The leaves of the plant used for an experiment for the process of photosynthesis should initially be placed in the dark for 24 to 48 hours to destarch the leaves.
During this period, all the stored starch will be used up by the leaves as the plant won’t be carrying out any photosynthesis. The leaves will thus not show the presence of starch.
