Subtractive hybridization:

  1. A technique called subtractive hybridization is employed to isolate a DNA fragment that was absent from a particular DNA sample.
  2. Let's say that the DNA for a certain gene was deleted, resulting in a genetic problem.
  3. This particular DNA segment will not be present in a sample of DNA from the chromosome of the affected DNA person.
  4. We must locate a healthy person and remove the DNA section in order to obtain the missing DNA.
  5. For instance, the Xp21 band, which lies at the center of the short or p-arm of the X chromosome, contains the DMD gene, a gene responsible for Duchenne muscular dystrophy.
  6. A patient's missing Xp21 band was discovered using light microscopy.
  7. In order to fix this, a healthy person was found from whom subtractive hybridization of the mutant chromosome with the healthy chromosome could be performed in order to clone the DMD gene.
