Tissue culture is a technique in which desired plants are grown under controlled nutritional and environmental conditions. It involves the following steps:
∙ The plant having the desired characteristics is selected.
∙ The most suitable part of the plant preferably the growing tip is excised out.
∙ The explant is throughly cleaned with water to remove impurities and then sterilised to remove microbial contamination. Thenafter, the explant is transfered to the nutrient media. The cultures are incubated so as to provide proper conditions like temperature, moisture, etc., for their growth and regeneration.
∙ Callus is formed. Further, it differentiates to form roots and shoots and eventually a complete plant.
∙ The plantlets are removed from the cultures and prepared for the soil transfer. This transfer to fields is known as hardening.
