Explain the structure and principle used in Transmission electron microscope?

In a Transmission Electron Microscope a beam of electrons is made to pass through the specimen. The thickness of the specimen is maintained to 10nm. The beam after passing through the specimen is focussed on an fluorescent screen, X-ray sheet etc to obtained a magnified image.
