PCR or polymerase chain reaction is a technique used in molecular biology to create several copies of a certain DNA segment.

PCR occurs in three steps:

- Denaturation: The high temperature melts the hydrogen bonds and separates the two strands of double-stranded DNA. The temperature is maintained at 92-96℃.

- Annealing: The two oligonucleotide primers anneal at desired sequences in the separated strands. It occurs at 55-65℃.

- Extension: Finally, Taq DNA polymerase forms the complementary strands by adding deoxyribonucleotides at free 3' OH end of the primers. It is carried out at 72℃.
