Dot blot

  1. Dot blot, also called slot blot is a technique used in molecular biology to detect proteins.
  2. It starts with spotting the RNA or DNA samples onto the nylon membrane.
  3. This nylon membrane is developed with a radioactive probe and then exposed to photographic films.
  4. Hybridization occurs between the single-stranded DNA or RNA strands with the single-stranded probes.
  5. The samples that contain RNA or DNA complementary to the given probe will give a black spot on the film.
  6. Various concentrations of the sample can be dotted in a side-by-side pattern on the membrane.
  7. This technique can also be used for protein detection using primary and secondary antibodies as shown below:
