How many below statements are incorrect with respect to various molecular biology techniques?
Agarose gel electrophoresis is the procedure employed for separating the mixture of DNA fragments which are formed on treating isolated DNA with restriction endonuclease enzymes. Thus statement 1 is incorrect.
Polymerase chain reaction (PCR) is a technique employed for amplifying a section of DNA (gene of interest). Taq DNA polymerase enzyme used during the process of PCR helps in binding the bases to the denatured DNA strand thus leading to the extension of DNA fragments. Thus, statement 2 is correct. But PCR is not a technique of DNA isolation, hence statement 4 is incorrect.
Restriction enzymes, cut DNA segments at a specific sequence. For example, EcoRI cleaves the DNA at specific sequence GAATTC, between the G and the A nucleotides. For this to happen the enzyme and the purified DNA are mixed and incubated at optimum conditions of temperature, pH etc. Thus, statement 3 is true.
During the process of electrophoresis, EtBr (Ethidium Bromide) gets intercalated between the bases of DNA which act as illuminating agent when subjected to UV radiation. Hence, statement 5 is correct.
