Agarose gel electrophoresis is a laboratory technique commonly used to separate charged particles (like DNA) of varying sizes in an agarose matrix under the influence of an electric field. The technique involves the following steps:-

  • Agarose powder is dissolved in TAE or TBE buffer. Ethidium bromide is also dissolved in it. It is casted into the tray.

  • Combs are inserted into the gel. It is then cooled for gel solidification.

  • After solidification, the combs are removed to obtain wells.

  • DNA samples are pipetted into the wells.

  • Buffer is added to the electrophoresis chamber.

  • Power is turned on to allow current conduction.

  • Once migration is done, the power supply is turned off.

  • The gel is procured out and viewed under UV light.

  • The part of the gel containing the gene of interest is identified and excised.

  • Elution of DNA is done to get the gene of interest.
