Agarose gel electrophoresis is a laboratory technique commonly used to separate charged particles (like DNA) of varying sizes in an agarose matrix under the influence of an electric field. The technique involves the following steps:-
Agarose powder is dissolved in TAE or TBE buffer. Ethidium bromide is also dissolved in it. It is casted into the tray.
Combs are inserted into the gel. It is then cooled for gel solidification.
After solidification, the combs are removed to obtain wells.
DNA samples are pipetted into the wells.
Buffer is added to the electrophoresis chamber.
Power is turned on to allow current conduction.
Once migration is done, the power supply is turned off.
The gel is procured out and viewed under UV light.
The part of the gel containing the gene of interest is identified and excised.
Elution of DNA is done to get the gene of interest.
