The cDNA made from proinsulin mRNA was used for gene transfer to produce insulin by Walter Gilbert and Lydia Villa-Komaroff from Harvard University. This gene was incorporated within the penicillinase gene site and the resultant product was a mixture of proinsulin and penicillinase. Gilbert used the enzyme trypsin to remove penicillinase portion of the mixture to derive active insulin.
