DNA Ligase - Joining the DNA fragments

Enzyme DNA ligase is used to join two DNA fragments by the formation of phosphodiester bonds. It is used in recombinant DNA technology to join the alien DNA with the plasmid.

Ti-plasmid - Agrobacterium

Agrobacterium tumefaciens is a bacterial plant pathogen which carries the Ti plasmid or tumour inducing plasmid (extrachromosomal DNA). The T-DNA segment of the Ti plasmid integrates itself into the plant cell’s genome to induce tumour formation in plants.

Taq polymerase - Thermostable DNA polymerase

Taq polymerase is a thermostable enzyme obtained from bacterium Thermus aquaticus. It is used in PCR technique to withstand variable temperature conditions.

Insertional inactivation - β-galactosidase

Insertional inactivation is the process of inactivation of a marker gene by the insertion of foreign gene in its coding sequence.

For example, insertion of a foreign gene in the coding sequence of β-galactosidase gene, synthesis a non-functional enzyme, which does not convert chromogenic substrate into a product. Hence only the non-recombinant colonies produce blue colour due to the presence of chromogenic substrate and absence of insert. In this way, it helps in the selection of recombinants.
