rDNA (Recombinant DNA) technology is the process of joining two different DNA segments in a host cell where genetic recombination occurs. Plasmid DNA is an extra chromosomal DNA which is a natural vector used to transfer the DNA segment containing the Gene of interest into the host cell.

During the process of gel electrophoresis DNA takes up sufficient amounts of Ethidium bromide (EtBr). This EtBr is intercalated between the DNA bases which in presence of ultraviolet light exhibits fluorescence.

Agarose gel electrophoresis (AGE) is a process of separating a mixture of charged molecules like DNA under the influence of electric current. When the mixture containing DNA is subjected to electrophoresis,DNA fragments move towards the positive electrode as DNA is negatively charged.

Agarose is a polysaccharide, generally extracted from certain red seaweed. It is a linear polymer made up of the repeating unit of agarobiose, which is a disaccharide made up of D-galactose and 3,6-anhydro-L-galactopyranose.
