A) For staining the onion peels, a dilute solution of safranin is used.
A minimum of 2 to 3 minutes of keeping the cell in contact with the staining agent is required after which, the excess is removed via blotting paper. Safranin is a staining agent used in histology and cytology to color the cells and visualize them under the microscope. Safranin reacts with lignin and stains it red. Therefore, only the lignified tissues such as xylem and sclerenchyma are turned red and all other tissues like parenchyma, collenchyma, and phloem remain colorless.
B) Methylene blue is used to stain the cheek cell taken out of the inside of the mouth.
