Dear Student, In vitro clonal propagation technique is used to produce virus-free healthy plant from a diseased sugarcane plant. The apical tissue or bud excised from the diseased plant is used to obtain virus free plant. Apical bud is meristematic in nature, which are actively dividing cells and always they are resistant to any infection. The use of meristematic tissue to produce virus free plant is known as meristem culture. In other words, Meristem culture is basically an in vitro culture of a dome shaped structure that measured less than 0.1mm in length of a plant. To obtain virus free healthy plants from a diseased one by tissue culture technique apical and axillary meristems of the diseased plant will be taken out.This is because they are generally free of infection or carry a very low concentration of the viruses.
