In order to extract DNA from a cell, firstly cell lysis or breaking of the cell membrane is done.

If the cell has a cell wall like plant, bacterial, or fungal cells, then additional enzymatic treatment is given to degrade the cell wall.

This includes cellulase treatment for plant cells (cell wall made of cellulose), lysozyme treatment for bacterial cells (cell wall made of peptidoglycan) and chitinase treatment for fungal cells (cell wall made of chitin).

Later protease, RNase treatment is given to digest macromolecules like proteins and RNA respectively, etc.

Finally, chilled ethanol is added to precipitate the DNA.

The purified DNA can be seen in the test tube as a collection of fine threads which can be scooped and swirled around a glass rod or wooden spooling stick. This process is called spooling. This helps to isolate the purified DNA threads from solutions.
