The DNA molecule pBR322 is a plasmid. It is an artificial cloning vector constructed by Bolivar and Rodriguez.

pBR322 has two antibiotic resistance genes.
Ampicillin resistance gene and tetracycline resistance gene.
These genes confer resistance against the antibiotics (ampicillin and tetracycline).

pBR322 contains several restriction sites which are specific DNA sequences at which specific restriction endonucleases bind and cleave phosphodiester bonds.
Pst I, Pvu I, EcoR I, Cla I, Hind III, BamH I, Sal I, Pvu II are some of the restriction/cloning sites found in pBR322.

BamH I site is present within the tetracycline resistance gene.
As the gene of interest (GOI) is inserted at the BamH I site, the tetracycline resistance gene will get inactivated. Hence, the bacteria into which this plasmid is introduced will not have resistance against the antibiotic tetracycline.

However, as the ampicillin resistance gene is functional in the plasmid, it provides resistance against ampicillin to bacteria.


The bacteria will not have resistance against the antibiotic streptomycin as there is no gene in the pBR322 that confers resistance against streptomycin.
