The correct option is A 1002
The first nucleotide at 5’ end contains three phosphate groups rest all has one phosphate group. During the formation of phosphodiester bond between successive nucleotides, the two phosphates are removed from the 5' end of the second nucleotide to provide energy for the bond formation. This does ot happen with the first nucleotide. See the diagram attached below.


So, for a 1000 nucleotide mRNA, there are 1002 phosphate groups.
