Replication in plasmid DNA is divided into 3 stages: initiation, elongation and termination. DNA polymerase cannot initiate de novo replication, so there is need of the independent generation of primer. This is solved in the plasmid. Plasmid coated initiation proteins catalyze initiation by recognising plasmid specific DNA sequence and determine the point from which replication starts. Plasmids do require host-encoded proteins for initiation of replication. Elongation is carried out by DNA Polymerase ||| with the participation of other host proteins. Termination of replication has specific requirements controlled by the host apparatus.
