The nucleus is composed of DNA, basic proteins, non-basic proteins, RNA, lipids and minerals. The most important basic proteins are the histone proteins which are required for coiling and packaging of DNA present in chromatin material. The histone proteins are rich in basic amino acids like lysine and arginine and hence have a positive charge naturally. Haematoxylin stains basophilic substances.
