When the bond formed between an enzyme and an inhibitor is a strong covalent bond, irreversible inhibition will occur. An irreversible enzyme inhibitor forms a strong covalent bond at the enzyme active site with amino acid side chain group. As the bond formed is a strong bond additional substrate addition does not reverse the inhibition process and the enzyme is permanently deactivated. Thus the body degrade the enzyme inhibitor complex and synthesis of new enzyme.
