DNA library:

  1. The DNA library is a collection of DNA fragments that have been cloned into vectors to recognize and isolate desired DNA fragments.
  2. There are two types of libraries – cDNA and genomic libraries.
  3. It requires the preparation of the DNA library and preparing the labeled probe.
  4. Gene libraries are stored as a bacterial culture of E.coli cells where each has a plasmid comprising a different insert.
  5. Culture is cultivated, diluted, and plated on different agar plates for colonies to space out from each other.
  6. Colonies are passed to nylon filters and DNA from each colony is liberated from cells by lysing it with detergent.
  7. Cellular elements are rinsed from filters.
  8. DNA adheres to nylon membranes and then is denatured forming single strands.
