There are two methods for sequencing the total DNA from a cell namely, ESTs (Expressed Sequence Tags) and sequence annotation.
In ESTs method, the part of a DNA is transcribed into mRNA and translated into proteins. Later, it sequences the part denoting a gene.
In the sequence annotation method, the total DNA from a cell is isolated and converted into smaller fragments and cloned in a suitable host using specialised vectors. The cloning results in the amplification of each piece of DNA fragment. Later, these fragments are sequenced using automated DNA sequencers, for the determination of amino acid sequences in proteins.
