Stanley Cohen and Herbert Boyer are considered to be the pioneers of recombinant DNA technology. They were the first people to use two plasmids (EcoR1 and pSC101) and recombine then with the help of restriction enzymes and DNA ligase. EcoR1 had resistance to kanamycin and pSC101 had resistance to tetracycline. Then the recombinant plasmid was transformed into a bacteria with the help of calcium ions and then plated on media containing both tetracycline and kanamycin. Some bacteria survived, which meant that the recombination was successful and this was hailed as an eureka moment in genetic engineering.
