You are synthesizing DNA in vitro and add DNA ligase from an old container to the mix by mistake. If the DNA ligase is still active, the reaction will work fine and you will obtain normal results. What would you expect to happen if the DNA ligase preparation is no longer active?

Dear student, During DNA replication, DNA ligase plays role in joining the Okazaki fragments of the lagging strand by providing the phosphodiester bond. So, if the DNA ligase preparation is no longer active the fragments of DNA won't be joined and a stable DNA molecule won't be obtained. Regards
