Can you explain why twelve molecules of water as substrate are used in the equation given above ?

The twelve molecules of water are used in the process of photosynthesis because for this process to occur it requires 12 NADPH molecules. Two molecules of NADPH require two water molecule to balance electron and proton in the reaction, so for the formation of 12 NADPH molecules, 12 water molecules are required.
