Principle of PCR :

  1. The polymerase chain reaction is a molecular biology technique through which the DNA of an organism can be amplified to make thousands or billions of copies of DNA of interest
  2. For amplifying the DNA, the target DNA is denatured at a high temperature to produce two single strands, two primers (short oligonucleotides having a complementary sequence to the target DNA) bind to the target DNA sequence and synthesize the new strand by adding deoxynucleotides by Taq polymerase enzyme.
  3. One cycle of PCR gives two copies of the target DNA.

Principle of DNA Sequencing :

  1. The process by which the nucleotide bases (Adenine, Guanine, Cytosine, and Thymine) along the DNA sequence are determined is called DNA sequencing.
  2. The two methods of DNA sequencing are Sanger sequencing and Maxam Gilbert sequencing.
