Antigen binding occurs at the N-terminus of the variable region of both light and heavy chains and the Fab fragment contains both these regions. The Fc fragment contains only the constant region of the heavy chains and it doesn't take part in the antigen binding. The enzyme papain (derived from papaya) breaks the antibody at the hinge region to produce these two fragments.

Variable regions of either of the heavy or light chains alone cannot take part in the antigen binding.

Immunoglobulin or antibody is a ‘Y’ shaped molecule, formed of amino acid sequences which specifically bind to the foreign antigens to disable them or mark them for destruction.

Conventional antibodies are formed of four polypeptide chains, two identical heavy chains and two identical light chains; light chains of short length and lower molecular weight and heavy chains of high molecular weight with longer length. Chemicals like reducing agents such as pepsin, papain, ficin etc. are used to fragment the antibodies by breaking the disulphide bonds within the hinge regions, for different research purposes to check the binding capacities of the antibodies.
